MicroRNA‑325 inhibits the proliferation and induces the apoptosis of T cell acute lymphoblastic leukemia cells in a BAG2‑dependent manner

The inhibitory effect of microRNA (miR)-325 in multiple different types cancer cell has been identified; however, its biological function T acute lymphoblastic leukemia (T-ALL) remains unknown. Moreover, Bcl-2-associated athanogene (BAG)2 is highly expressed a various tumors and regarded as an anti-apoptotic gene. In the present study, roles miR-325 BAG2 T-ALL line (Jurkat cells) were investigated. expression levels clinical blood samples from healthy donors pediatric patients with T-ALL, well lines was detected using western blot analysis and/or reverse transcription-quantitative PCR. Dual-luciferase reporter gene assays TargetScan used to evaluate interaction between miR-325. Small interfering RNA technology applied knockdown Jurkat cells. effects mimic downregulation on proliferation apoptosis assessed by MTT assay, flow cytometry analysis. results revealed that downregulated lines, lowest exhibited opposite results. markedly induced inhibited addition, overexpression significantly growth promoted cells, these being eliminated overexpression. conclusion, findings study demonstrated directly targets introduction can accelerate suppress cells silencing expression.
